JAMM Therapeutics is a pharmaceutical company that is focused on finding small molecule modulators to treat systemic lupus erythematosus (SLE). Type I interferons (IFNs) are a family of cytokines with antiviral, antiproliferative, and immunomodulatory abilities. They work by attaching to a receptor complex composed of the IFNAR1 and IFNAR2 subunits. IFNAR1 polyubiquitination is one of the processes that control type I IFN activation. This process is reversed by a cytoplasmic complex called DUB (BRCC36 isopeptidase complex), which preserves IFNAR1 and enhances IFN-dependent reactions. In both humans and mice, type I IFNs are essential in the development of SLE. In BRISC-deficient mice, the mice present improved clinical functioning, lowered IFN responses, and protection from immunopathology associated with IFN.
